Application【应用】	碱性磷酸酶可与抗体及其他蛋白质偶联而用于ELISA、免疫印迹和组织化学检测。它通常用于使蛋白质和核酸去磷酸化。当需要高灵敏度时，可用于蛋白标记。碱性磷酸酶也可用于使DNA或RNA的5′-末端去磷酸化以防止自连。在用碱性磷酸酶去磷酸化后，DNA或RNA也可用放射性标记的磷酸盐(通过T4多核苷酸激酶)进行标记。产物 P8361 已被用于去磷酸化 Dishevelled (Dvl)，这是 Wnt/β-连环蛋白信号通路中的关键组分。使用该 ALP 酶开发了一项试验，以评估两种蛋白 CRP-1 和 CRP-2 的冷冻保护活性。该酶已用于抑制前列腺酸性磷酸酶的环核苷酸类似物的抑制研究(使用荧光试验)。它在检测系统中常用作“报告者”，即与特异性识别靶分子的蛋白(抗体、链霉亲和素等)偶联。碱性磷酸酶也被用于去磷酸化酪蛋白。本产品在发酵或纯化过程中未使用任何动物源材料。
Biochem/physiol Actions【生化/生理作用】	碱性磷酸酶使蛋白质和核酸去磷酸化。碱性磷酸酶是由两个相等的亚单位组成的二聚体。这种酶是一种糖蛋白，含约 12% 的碳水化合物(6% 己糖和 6% 其他中性糖)。每个碱性磷酸酶分子含有四个锌原子和四个二硫键。在镁存在的情况下，碱性磷酸酶活性达到最大。对磷酸对硝基苯基磷酸酯、磷酸苯酯、磷酸酚酞、α-甘油磷酸酯、β-甘油磷酸酯、2-磷酸甘油酯、磷酸丙糖酯、葡萄糖6-磷酸、葡萄糖 1-磷酸、果糖 1-磷酸、果糖 6-磷酸、腺苷 5-磷酸腺苷 3-磷酸、磷酸烯醇丙酮酸和 β-烟酰胺腺嘌呤二核苷酸磷酸等磷酸单酯的水解进行催化。砷酸盐、半胱氨酸、碘、无机磷酸盐、焦磷酸盐、磷酸二异丙酯、三苯基磷酸盐、氟磷酸二异丙酯和 L-苯丙氨酸是碱性磷酸酶的一些强抑制剂。该酶在 pH 7.5-9.5.2 范围内最稳定，酶活性的最适 pH 为 pH 8-10。
